We are searching for a Senior Scientist within Immuno-Oncology, Discovery Oncology Flow Cytometry Core Facility to be in Spring House, PA and have the following responsibilities:
Proactively support Oncology Discovery and Translational Research by providing flow cytometry expertise working within a core facility.
Maintain 15-20 flow cytometers in a multifaceted industry setting. Monitor all calibration results to ensure optimal performance and maintain appropriate instrument configurations. Create/update SOPs for core instrumentation and staining protocols.
Perform all preventative maintenance activities for instruments and computers, including weekly/monthly maintenance procedures, scheduling service appointments, database back-up, quarterly data transfer, collaboration with IT for computer upgrades, and maintaining network and data share accessibility.
Aid users in technical troubleshooting to minimize instrument downtime. Resolve common issues independently, communicate issues to users and update reservation calendars as required.
Process tissue (blood, lymph nodes, tumor, prostate, lung, muscle, spleen, and thymus) to single cell suspension for analysis of immune, tumor, and/or stromal cells by Flow cytometry or other biological methods to support Oncology efforts
Stain and characterize both primary immune cell populations and cell lines by multiparameter (8-40+ colors) flow cytometry, including intracellular and cytokine staining and cell stimulation techniques.
Perform FACS (cell sorting) as a scheduled service of the core facility. Responsibilities include appointment consultation, instrumentation setup and full operation of the cell sorters including technical troubleshooting. Provide data reports to the end user in a timely manner.
Train all new users on flow core protocols and procedures including basic start-up, experimental setup, compensation/unmixing and shutdown procedures. Lead small group hands-on training sessions on instrumentation.
Support all groups with advanced spectral assay/panel design, unmixing set-up, and data analysis using FlowJo and OMIQ platforms applying high dimensional data reduction and visualization techniques.
Maintain detailed and organized experimental records and stay compliant on all relevant internal and external training requirements.
Strong leadership and mentoring abilities, with a passion for developing talent and encouraging a culture of scientific excellence.
Communicate research findings and recommendations effectively through written reports, presentations, and scientific publications.
Remain current on the latest flow cytometry trends, assays, and technology and reagents to share across the department. Identify emerging trends and opportunities in the field and provide strategic recommendations to senior management.
Ability to travel to attend specific trainings / conferences as needed up to 10% of time.
Qualifications:
Ph.D. in a relevant scientific subject area (e.g., Chemistry, Cell Biology, Pharmacology, Biochemistry) with a minimum of 2 years of industry or academic experience within a flow cytometry core facility. Alternatively, Masters w/ 4-6 years of relevant experience or Bachelor’s w/ 7-9 years of relevant experience.
At least 1 year experience with full spectral high parameter multi-color flow cytometry (25+ colors) including panel design best practices, spectral unmixing, fluorochrome compatibility, and data QC is required.
Experience with technical troubleshooting on conventional and spectral instrumentation and corresponding software from vendors including BD is required. Cytek, or Sartorious, or SONY experience is highly desired.
Demonstrated proficiency with high parameter (25+ color) panel design, assay optimization, qualification and assay transfer procedures for the evaluation of clinical trial samples is required.
Independent operation of FACS (cell sorting) instrumentation on BD platforms and basic troubleshooting is required.
Demonstrated strong leadership and mentoring abilities to train new users on the theory and practical application of flow cytometry is required.
Experience in design and interpretation of cell-based assays/experiments to interrogate questions in immunology and oncology is highly desired.
Experience with aseptic technique, cell culture and basic immunogenicity assays is preferred.
Experience with in vivo models of tumor biology, tissue processing, and subsequent flow cytometry workflows is preferred
Excellent and timely communication/collaboration skills are required.
